Detection of rice GM events containing p35S::bar sequences

In line with Commission Decision 2006/578/EC of 23 August 2006 on emergency measures regarding the non-authorised genetically modified organism LL RICE 601 in rice products the following documents are provided here:

  • "Grain testing method for detection of rice GM events containing p35S::bar sequences using RT-PCR protocols PGS0494 and PGS0476", developed by Bayer CropScience.
  • "Verification of the Bayer CropScience 35S-BAR method for the detection of LL62/LL601 rice using Real-Time PCR", produced by the US Department of Agriculture.

 

These documents have been evaluated by the Joint Research Centre as European Union Reference Laboratory for GM Food and Feed (EURL GMFF) on the basis of the information available to the EURL GMFF as of 31 August. The outcome of this evaluation as given in the attached publication (" Report on the verification of a construct-specific detection method for identification of rice GM-events containing P35S::BAR using a real-time PCR assay" ) reflects the situation as of that date. The EURL GMFF will further investigate LLRICE601 particularly with respect to its molecular structure and to the possible occurrence of false positive and false negative test results. It will keep the dossier under constant review and publish here any updates to the methods and other relevant information as soon as the need arises. In these circumstances, it cannot assume any responsibility, in contract or in tort or on any other basis, for any consequences whatsoever which may ensue from the use of the methods as published herein.

Important note

In relation to the P35S::bar construct-specific method for detection of LLRICE601 and LLRICE62 events, the European Union Reference Laboratory for GM Food and Feed draws the attention to the following:

  • The EURL GMFF has shown that the method reacts positively with genomic DNAs from other GM-events (e.g. Bt176 maize, LLCotton25, Ms8 Rf3 oilseed rape), including some authorized ones. When thus tested on complex food and feed matrixes this could result in the generation of false-positive signals with respect to LLRICE601 or LLRICE62. Details can be found in the EURL GMFF verification report.
  • The EURL GMFF has observed that the amplicon size differs between the LLRICE601 and the LLRICE62 events; the LLRICE62 amplicon being the smallest and measuring around 100bp.
  • The EURL GMFF recommends routine testing of the negative control samples for the absence of LLrice601 and LLRice62 events.
